 AB    OBJECTIVE: The capability of CD34+ SC from HIV seropositive hemophiliacs
       to differentiate into mature and functional T cells when cocultured in
       allogeneic cultured thymic epithelial fragments (CTEF) was evaluated.
       METHODS: Bone marrow CD34+ SC, obtained from 12 HIV seropositive
       hemophiliacs by E-rosette depletion, SBA depletion, adherence and CD34
       panning, were > 95% CD34+ and CD2-CD3-CD4-CD8-. CTEF were obtained from
       thymus glands, depleted of thymocytes and dendritic cells by
       2'-deoxyguanosine, and cultured for 2-14 days prior to seeding with
       10(5) CD34+ SC. The CD34+ SC were analyzed for HIV proviral DNA by PCR
       using pol, gag and env probes and found to be infected in 1/12. RESULTS:
       Phentoypic analysis of the SC-CTEF cocultures at 18 days revealed T cell
       differentiation of SC: CD7 46.1%, CD2 34.9%, CD3 44.7%, CD4 23.3%, CD8
       13.2%, TCRab 40.7%, TCRgd 5.2%. T cells were of SC origin determined by
       HLA-A typing and labelling of the SC with a fluorescent intracellular
       probe, CMTMR. Lymphoproliferation of the SC-CTEF T cells to PHA
       stimulation revealed 20,932 cpm (SI 5.9) with autologous B cells and
       8,552 cpm (SI 2.4) with CTEF B cells (p < 0.05). MLC responses by
       SC-CTEF T cells revealed no reactivity to CTEF B cells. CONCLUSIONS:
       These studies revealed that HIV seropositive hemophiliacs' CD34+ SC were
       rarely HIV infected, were capable of differentiating into mature and
       functional T cells when cocultured in allogeneic CTEF, and were tolerant
       to CTEF HLA antigens.
