 AB    OBJECTIVE: The V3-loop of HIV-1 bears principal neutralizing determinant
       (PND). We have investigated anti-PND antibodies (Abs) with a total of
       170 (93 seropositive and 77 seronegative) Japanese hemophilia A using
       synthetic peptides. Several patients were also analysed on cDNA
       sequences of V3-loop. METHODS: Anti-PND Abs were measured by an
       enzyme-linked immunosorbent assay (ELISA) using 7 synthetic peptides
       (GPGR-motif: IIIB, MN, RF, SC, and WMJ-2, GLGQ- or GPGQ-motif: Af1.Con
       and Af.2 Con), each 15 amino acid residues deduced from cDNA sequences
       of the V3 mutants. Each specific Ab binding was determined after
       subtraction of the non-specific from the total. Arbitrary anti-PND Ab
       titer was also determined by limited dilution of serum, in which below
       the titer 20 indicates the Ab negative. RESULTS AND CONCLUSIONS: The
       serum specimens with HIV-1 Ab negative uniformely indicated below the
       cut-off value (0.254 at OD405 nm) on anti-PND Abs. Positive anti-PND Abs
       were found in the 93 sera with HIV-1 Ab positive in the following
       frequencies; 3 for IIIB, 43 for MN, 2 for RF, 0 for SC, 0 for WMJ-2, 1
       for Af.1 Con, and 4 for Af.2 Con. Six out of 7 AIDS-patients had the
       anti-MN Ab below 20, whereas one had the titer 100. One AC-patient with
       high anti-MN Ab titer (200-600) indicated no significant decrease of
       CD4(+) cell count over the last 8 years. cDNA sequence analysis of the
       V3-loop in HIV-1 isolated from patient sera also indicated no major
       amino acid substitution around PND in the patients with high anti-MN Ab,
       whereas those with low anti-MN Ab had few amino acid substitutions
       around PND.
