AB Particular strains of HIV may dominate the CNS of infected individuals
and it is possible that the HIV Nef protein plays a role in
differentiating CNS and peripheral blood isolates. To address this, 6
pairs of coincident isolates from the CSF and peripheral blood of
patients with AIDS or ARC were analysed for their ability to replicate
in primary blood mononuclear cells and primary macrophages. Peripheral
blood isolates appeared to have enhanced replication in all the cell
types examined but the replicative abilities of paired isolates were
very similar. CSF isolates were less likely to induce CPE and syncytia
in PBMC and macrophage cultures. Isolates were examined for their
ability to express the Nef protein by flow cytometry and immunoblotting
and the nef gene of isolates was PCR amplified and sequenced. Most
isolates appeared to express Nef and sequence analysis revealed little
difference in nef between CSF and blood isolates. No differences between
Nef of CSF and peripheral blood isolates were apparent by parameters
       used in this study.
