 AB    The kinetics of individual viral RNA synthesis of HIV-1 regulatory and
       structural genes in ACH2 cells after PMA induction were analysed at the
       single cell level using in situ hybridisation with splice-junction
       probes. Before PMA stimulation, only a small fraction (< 23%) of the
       cells expressed tat, nef, env or gag/genomic RNA, no cells expressed
       rev. One hour after induction, the percentage of tat, nef, env and
       gag/genomic RNA expressing cells increased to 85%, 99%, 49% and 74%,
       respectively, only 12% of the cells expressed rev mRNA. Seven hours
       after induction, 96% of the cells expressed tat mRNA; 100% expressed env
       and gag/genomic RNA; 38% expressed rev mRNA. Thereafter the expression
       level of env and gag/genomic RNA, but not tat and rev mRNA, on
       individual cells continued to increase. Our results showed that in the
       PMA-ACH-2 system, induced expression of HIV-I env and gag/genomic RNA
       followed the enhanced expression of tat and nef mRNA but preceded the
       induced expression of rev mRNA.
